Not surprisingly, there is no hint as to who might portray the new Spider-Man - also, Bruce Banner/Hulk wasn't confirmed yet.

However, Ant-Man was now confirmed, as well as a character which has appeared in The Incredible HulkWilliam Hurt as General Thaddeus "Thunderbolt" Ross will also come into play, thus reconnecting the only Hulk solo film released by Marvel to the MCU.

Captain America: Civil War synopsis

Captain America: Civil War picks up where Avengers: Age of Ultron left off, as Steve Rogers leads the new team of Avengers in their continued efforts to safeguard humanity. After another international incident involving the Avengers results in collateral damage, political pressure mounts to install a system of accountability and a governing body to determine when to enlist the services of the team. The new status quo fractures the Avengers while they try to protect the world from a new and nefarious villain (via Comic Book Movie).

Clearly, this new villain is Zemo, who apparently has a much bigger role than previously thought. Furthermore, we do believe that his machinations are, ultimately, the ones which will set Captain America and Iron Man on a collision course - in a way this suggests that, until the third act of Civil War, Zemo will be taken out of the picture.
